研究工作的特色和意义

自噬作用是广泛存在于真核细胞内的一种溶酶体依赖性降解途径,它通过对蛋白质和受损的细胞器的吞噬降解,是细胞内的再循环系统和细胞进行自我保护的一种重要机制,在维持细胞存活、更新、物质再利用和内环境稳定中起着重要作用。自噬的过度表达或失活与2型糖尿病密切相关。我们以NLRP3-ASC-Caspase 1炎性小体与自噬二者间的相互作用的级联效应作为研究切入点,采用细胞应激与代偿等细胞生物学技术,质粒转染、RNA干扰及Western Blot等其他分子生物学技术,以及糖尿病动物模型,利用代谢组学、蛋白质组学等系统生物学研究手段,并结合糖代谢病理生型糖尿病发生发展过程中自噬参与NLRP3依赖性慢性炎症引起的胰岛素抵抗及2型糖尿病的分子机制,相关研究成果已发表在Diabetologia, JBC 2013, Cell death & disease 2014) 等高水平代谢杂志上。将产生具有自主知识产权,研究水平达到或接近国际水平的分子内分泌代谢学实验室。
